Chemistry

The Lokey Lab focuses on the synthesis and study of cyclic peptides and peptidomimetics, with an emphasis on understanding and optimizing membrane permeability and oral bioavailability. Our chemistry spans solid-phase and solution-phase peptide synthesis, protecting group strategies, N-alkylation methods, and library design.

Subsections

  • Solid-Phase Synthesis — Protocols for on-resin peptide assembly, including coupling reactions, N-methylation, esterification, and peptoid methods
  • Solution-Phase Methods — Cyclization, amide coupling with DSC, and other solution-phase transformations
  • Protecting Groups — Reference for amine and carboxylic acid protecting groups used in peptide synthesis
  • N-Alkylation of Boc Amino Acids — Procedure for N-methylation (and trideuteromethylation) of Boc-protected amino acids
  • Design Rules & Library Design — Guidelines for designing cyclic peptide libraries, sequencing, chromatography, and isotopic labeling
